Alex made it 2-0 in the 39th minute with a perfectly placed free-kick.
The Russian Premier League’s top scorer Welliton got on the end of a cross seven minutes after the break for Spartak’s thrd goal.
Spartak, who were beaten 5-2 by Alaniya earlier in the season, are in fourth place in the Premier League, six points behind third placed CSKA, who have played two games less.
While the Moscow club’s domestic form has been indifferent this season, they, like Chelsea, have a 100% record in the Champions League after two games.
Chelsea may be without Didier Drogba, who has a fever, for Tuesday’s clash.
