The German youngster covered the 181.4-kilometer flat stage in four hours and 38 minutes, beating second-placed Australian Allan Davis of GreenEDGE and Team Sky's third-placed Ben Swift in a sprint finish.
Spanish cyclist Jonathan Castroviejo remains the leader after he took the red jersey Saturday when his Movistar team won the opening team time trial in Pamplona.
Defending champion Juan Jose Cobo is four seconds behind his teammate Castroviejo.
On Monday, the peloton will face its first venture into the mountains, a 155 kilometer stage with the finish at Eibar.
The Vuelta consists of 21 stages and finishes September 9 in Madrid.
